You can CRB a DU mutation and it will not effect a lion scrotum.

To do this:
1.) Breed your DU (Do not use any items)

2.)CRB that litter then wait 3 days for her to be in heat.

Note you can follow from this point 3a or 3b I recomend following 3a

3a.) Use a lion scrotum on your DU female, and any other items like a Gnawrock, Great tit, GoP, or Buffalo scrotum.

3.5a.) Send your stud request with the above items, or use the items on your king if you plan to breed her to him.

Note there is not need to breed her twice unless you want to use double the items(I will explain is 3b). So from this point just nest her and wait 3 days for the birth or IBF her to see the cubs imediantly. If you use a buffalo scrotum she will have a minimum of 6 cubs and a max of 8 even without breeding her twice.

3b.) Use a lion scrotum and the items you want to use on the first stud. You must use either a GoP or buffalo scrotum during this first breeding as this determines how many cubs she has. You do not need to use a GoP or Buffalo Scrotum in the 2nd breeding.

3.5b.) After the first being is complete if you want half of the cubs to be from a second stud the CRB will not affect half of the litter and you must re use a lion Scrotum for the 2nd stud. This is not recomended as it is a waste of items in my opinion.

4b.) Nest lioness after she is bred to both studs then IBF or wait 3 days for the cubs.

Future referance:

CRB takes up its own slot if you use a CRB you can use that item along with a Lion Scrote, GMO, or Lion Meat.

Note you can not use these items together in a same breeding you many only choose 1 per breeding: Lion Scrote, GMO, or Lion Meat.

Grain of Paradise and Buffalo Scrotums take a different slot but you may only choose one of the two per breeding.

I don't remember about the CRB or lion scrot, but using a Buffalo Scrot will instead guarantee 6 to 8 cubs.

you need to use a CBR before you use a lion scrotum, and i was told that you only need 1 buffy scrote and it will activate for both breedings.
so youll get 6 cubs (if im right not sure tho)
